Various chemical laboratory reagents are used in hematology tests that are used to perform various lab tests in the laboratory. Most of them are commercially available in the market or can be taken by manufacture. A lab technician or lab technologist patent must have knowledge about the composition of different reagents in the laboratory and their preparation and maintenance hare are some regent and their compost ion.

Composition of  reagents


Drab kin solution (used in hemoglobin estimation)

  • Potassium ferricyanide: 0.2 gm
  • Potassium cyanide: 0.05 gm
  • Dihydrogen potassium phosphate: 0.14 gm
  • Distilled water: 1000 ml

Drabkin’s fluid should be clear and pale yellow having a pH of 7.0-7.4.

Turk’s fluid (WBC Diluting fluid)

  • Glacial acetic acid: 3.0 ml
  • 1% aqueous gentian violet: 2.0 ml
  • Distilled water: 195 ml

Hayem’s fluid (RBC Diluting fluid)

  • Mercuric chloride: 0.25 g
  • Sodium chloride: 0.5 g
  • Sodium sulfate: 2.5 g
  • Distilled water: 100 ml

Dacie’s fluid (RBC Diluting fluid)

  • 40% Formaldehyde: 5 ml
  • 3% Trisodium citrate: 495 ml

Diluting fluid (Platelets Diluting fluid)

  • 1% ammonium oxalate
  • Ammonium oxalate: 1g
  • Distilled water: 100 ml

Filter the solution and keep it in a refrigerator at 4 degrees Centigrade.

1% new methylene blue (staining solution for reticulocytes)

  • New methylene blue: 1.0 g
  • Iso-osmotic phosphate buffer (pH 7.4): 100 ml
  • Keep in a dark bottle and filter the solution before use.

Semen diluting fluid ( for sperm count)

  • Sodium bicarbonate 5 gm
  • Formalin (neutral) 1 ml
  • Distilled water 100 ml

Another solution will be added after some time
